Waifu is a memetic reference to Azumanga Daioh's Kimura-Sensei who has a thing for his female students, said students find a picture of a woman in his classroom who he refers to as "my waifu" or "my wife."

Mai Waifu is a meme, usually referring to a female anime character who a lonely male is pretending is their wife or girlfriend.

"Would waifu" is just an alteration of the meme simply saying you'd pretend to be married to the woman you're referring to.
